پاسخ ریشه‌زائی و برخی صفات بیوشیمیایی قلمه زیتون (Olea europaea L.) رقم کنسروالیا به اسید ایندول بوتیریک و پوترسین

چکیده

به منظور بررسی اثر غلظت‌های مختلف اسید ایندول بوتیریک (0، 2000 و 4000 قسمت در میلیون) و پوترسین (0، 100 و 200 قسمت در میلیون) بر برخی از صفات رویشی و بیوشیمیایی قلمه سخت ریشه‌زای زیتون رقم کنسروالیا آزمایشی به صورت فاکتوریل در قالب طرح بلوک‌های کامل تصادفی با سه تکرار اجرا شد. تجزیه واریانس داده‌ها نشان داد که اثر متقابل اسید ایندول بوتیریک × پوترسین بر درصد ریشه‌زایی، درصد کالوس‌زایی، تعداد ریشه، وزن ریشه، تعداد شاخه و وزن شاخه و همچنین بر میزان کربوهیدرات، میزان نیتروژن، نسبت کربن : نیتروژن (C:N)، آنزیم پراکسیداز و پلی‌فنل اکسیداز موثر بود. قلمه‌های تیمار شده با غلظت 4000 قسمت در میلیون اسید ایندول بوتیریک بیشترین میزان آنزیم پراکسیداز (114/0 میکرومول بر دقیقه بر گرم وزن تر)، پلی‌فنل اکسیداز (140/0 میکرومول بر دقیقه بر گرم وزن تر) و همچنین بالاترین درصد ریشه‌زایی را (100%) داشتند. در حالی که قلمه‌های تیمار شده با غلظت 4000 قسمت در میلیون اسید ایندول بوتیریک + 100 قسمت در میلیون پوترسین 80 درصد ریشه‌زایی داشت. با توجه نتایج این پژوهش، برای افزایش ریشه‌دهی در قلمه سخت ریشه‌زای زیتون رقم کنسروالیای می‌توان از غلظت 4000 قسمت در میلیون اسید ایندول بوتیریک یا از کاربرد توام 4000 قسمت در میلیون اسید ایندول بوتیریک به همراه پوترسین 100 قسمت در میلیون استفاده کرد.

This experiment was conducted to investigate the effect of different concentrations of indole butric acid (IBA) (0, 2000 and 4000 ppm) and puterscine (0, 100 and 200 ppm) on some of the morphological and biochemical traits of hard- rooting cutting of olive cv. Concervolia. Experimental treatments were arranged as factorial in randomized complete block design with three replications. Analysis of variance showed that IBA × puterscine effect was significant on rooting and callusing percentage, root number, root weight, shoot number and shoot weight as well as on carbohydrates and nitrogen content, C:N ratio, peroxidase and polyphenol oxidizes. The highest peroxides (0.114 µmol min-1 gFW-1), polyphenol oxidase (0.140 µmol min-1 gFW-1) and rooting percent (100%) were obtained in cuttings treated with 4000 ppm IBA, while rooting was 80 percentage in cutting treated with 4000 ppm IBA+100 ppm puterscine. Based on the results of this research, it can be cancluded that the concentration of 4000 ppm of indole butyric acid or the combination of 4000 ppm of indole-butyric acid and 100 ppm puterscine can be used for enhancing rooting in the hard- rooting cuttings of olive cv. Concervolia.
